Biosíntesis de poliuronidos en Phycomyces blakesleanus: caracterización de la glucuronosil transferasa

Extractos libres de celulas de Phycomycl'-b bla.kl'-6ll'-eanu6 presentaron
actividad de glucuronosil transferasa. La mayor parte de esta actividad
se encontro en forma particu l ada en una fracci6n mixta de membranas
(FMM) que sedimento a 54,000 x g. La adicion de acido mucorico o
mucorana a la fraccion membranal incremento la actividad enzimatica.
Por otro lado, la presencia de detergentes· no i6nicos tambiin l a
estimulo. Sin embargo, los detergentes i6nicos la inhibieron. Cuando se trato la fraccion mixta de membranas con detergentes no ionicos y
luego se recupero el sedimento por centrifugation , este presento
actividad enzimatica y ademas se hizo dependiente de la presencia de
los aceptores exogenos, acido mucorico y mucorana, siendo el primero
el mas efectivo. La temperatura y el pH optimos de la enzima fueron
de 22°C y 7.5 respectivamente en regulador de Pipes 22 mM. La enzi ma
se activo con los cationes divalentes Mn++> Fe++> co++> cu++> Mg++>
ca++ a una concen tracion de 8 mM. La Km para UDP-acido glucuronico
fue de D.3 mM. Tambien se estudio el efecto de azucares, nucleotidos
y azucares nucleotidos sobre la actividad enzimatica.
